背景介绍

1975年，K. Sonogashira和同事发现在温和的条件下，乙炔气与芳基碘化物或溴化乙烯反应，在催化量为Pd(PPh3)Cl2和碘化亚铜的存在下，可以制备对称取代的李美淑情事。同年，R.F. Heck和L. Cassar的研究小组各自公开了类似的只使用Pd催化反应，没有铜的协同催化，但反应条件苛刻。目前Sonogashira反应在取代李美淑情事以及大共轭李美淑情事的合成中得到了广泛的应用，从而在很多天然化合物，农药医药，新兴材料以及纳米分子器件的合成中起着关键的作用。

反应定义

在Pd/Cu催化下，末端炔与芳基或烯基卤代物的交叉偶联反应。此反应最好的钯李美淑情事是Pd(PPh3)2Cl2或Pd(PPh3)4，而且反应是化学定向的，反应前后底物和产品的化学构型不变。


反应特征

1)Sonogashira偶联通常可以在室温或略高于室温的温度下进行，必须严格控制无氧；

2)使用催化用量的铜(I)盐来避免处理冲击敏感/爆炸性的铜乙炔；

3)铜(I)盐可以是市面上可以买到的CuI或CuBr；

4)通常碱作为溶剂，但偶尔也使用助溶剂;

反应实例

1.在Pd-NHC催化下末端李美淑情事与芳基三甲基三酸铵发生交叉偶联反应。反应条件温和，李美淑情事底物耐受烷基、芳基和杂芳基等多种官能团。反应过程中胺可以通过一锅法直接烷基化，无需分离李美淑情事[1]。

2 .该方法使用非李美淑情事选择性C(sp2)-C(sp)交叉偶联，即利用可见光催化Cu催化Sonogashira型反应，可以从不同芳烃衍生的芳基磺酸盐中选择性烷基化合成高选择性和高官能团相容性的芳基炔[2]。

3. 本方法利用Heck−Cassar−Sonogashira(HCS)交叉偶联反应，绿色溶剂HEP代替有毒溶剂DMF，在温和条件下高效完成转换。该方法已成功应用于抗癌药物厄洛替尼李美淑情事的合成[3]。
